The aim of the project is to use the methods and experimental systems of optical tweezers, optical scalpel, and automated cytometer, that were developed using the finantial support of the previous scientific projects, for the study of the mechanical properties and arrangement of the interphase chromatin in the nuclei of living and fixed human cells. Theoretical and experimental experiences of the interdisciplinary team of applicants will be used for: - fluorescence labelling of chromatin in living cells, - laser induced fusion of labelled cells, - topologic analyses of the dye distribution inside living and fixed cells, - insertion of dyed nanoparticle into the living cell nucleus, optical confinement of this probe and detection of the region which is not accesible for the probe, - study of the mechanical properties of the chromatin using optical trapping of suitable probes.
