Procedure for the selective inhibition of some flower pigment biosynthesis steps in selected ornamental plants is developed with the use of molecular biology methods. Agrobacterium tumefaciens transformation of Dendranthema grandiflora and Rhododendron catawbiense is proposed, with "sense" and "antisense" constructs from anthocyanin synthesis genes chs, chi or dfr. Flower pigment variant plants obtained will be characterized by quick and reliable DNA-fingerprinting method that is developed in parallel research, on the collection of cultivars of black poplar at first. Microsatellites will be identified and sequenced in genome fragments enriched in 3b and 4b repeated sequences on streptavidine columns with biotin-bound complementary oligos. Useful microsatellite markers are selected and combined to develop standardised multiplex procedure for the complete identification in one amplification reaction. The same approach will be used for collections of other plant species. |
